Single Cell service

Single-cell transcriptomics – Single Cell RNA-Seq

The GENOM'IC core facility helps you explore the cellular heterogeneity of your samples using single-cell transcriptomics approaches.

We use Chromium technology (10X Genomics) for cell processing and library preparation. This method allows the transcriptome of thousands of individual cells to be analysed in parallel from a single sample.

Our service includes:

🔹 Initial quality control of the cell suspension or nuclei (viability, concentration)
🔹 Encapsulation of cells/nuclei on Chromium iX and preparation of libraries
🔹 Sequencing of libraries on NextSeq (Illumina), in paired reads
🔹 Quality control of raw data

Each project isdiscussed in advance to ensure optimal compatibility with the technical constraints of the single-cell approach (number of cells, integrity, sample type, etc.), and define best protocols and approaches. A personalised quote is provided based on your objectives.

Please do not hesitate to contact us directly with any project requests (u1016-genomique@inserm.fr).

Description of the Single Cell service

The GENOM'IC core facility offers expert support for your single-cell transcriptomics projects, using cutting-edge technologies developed by 10X Genomics.

We are currently one of the only platforms in the Paris region to offer personalised support at every stage of the workflow, from feasibility assessment to the production of usable data.

We support a wide range of 10X protocols, including:

🔹 3' scRNAseq (+/- surface proteins)
🔹 5' scRNAseq (+/- VDJ ou +/- surface proteins)
🔹 Multiome : expression + chromatin accessibility (ATAC) (nuclei only)
🔹 Flex GEM-X (+/- surface proteins) : new generation of kits for fixed or FFPE samples (human and mouse only)
🔹 Other specific protocols upon request

Before submitting a project, a preparatory meeting with the team is mandatory.

As each single cell project is unique, we help you define the optimal conditions:

  • Sample type and upstream preparation
  • Protocol adapted to scientific objectives
  • Number of cells, sequencing depth, desired analyses

We are available at every stage of the project to answer your technical questions and advise you. Contact us at the earliest stages of your project to discuss it.
