Cellules dendritiques, immunostimulation du microenvironnement viral et tumoral

Notre équipe analyse l’implication des cellules dendritiques (DC), des monocytes/macrophages et de cytokines dans les réservoirs du VIH et dans les réponses immunitaires antivirales et antitumorales. Elle étudie en particulier l’effet immunosuppresseur de l’enzyme IL-4I1 dans le mélanome.
Les cellules dendritiques (DC) sont les seules cellules présentatrices de l’antigène capables d'initier les réponses immunes adaptatives, tout en intégrant les signaux du microenvironnement tissulaire pour polariser ces réponses et effectuer des réponses d'immunité innée.  Nous avons mis en évidence la présentation croisée d'antigènes à partir de cellules vivantes par différentes populations de DC humaines, et en étudions les mécanismes pour lutter contre les métastases tumorales ou les réservoirs du VIH-1.
Nous avons participé à l’identification de réservoirs myéloïdes du VIH-1 avec Morgane Bomsel. Nous étudions les caractéristiques des réservoirs viraux et développons des approches expérimentales en vue de leur éradication.
Nous explorons le rôle des réponses IFN des DC et autres cellules myéloïdes contre VIH-1, VIH-2 et SARS-Cov-2 dans des cohortes de patients, contre SIV, ainsi que dans diverses situations physiologiques.
Nous avons identifié une nouvelle fonction de l’
IL-7 qui, produite par les muqueuses en phase aiguë d’infections virales et bactériennes, participe au recrutement des cellules immunitaires, initiant la réponse immunitaire. Nous étudions son rôle potentiel comme adjuvant de vaccination muqueuse.

Nous effectuons la caractérisation moléculaire des variants du JCV, virus opportuniste responsable de la leucoencéphalopathie multifocale progressive.
Nous avons découvert un rôle original de la phénylalanine oxydase  IL4I1 (IL4 induced gene-1), nouvelle cible thérapeutique dans le traitement du cancer, sur la biologie des lymphocytes B. Les mécanismes d'action de cette enzyme immunosuppressive sont étudiés dans le cadre du mélanome, dans un modèle murin et chez des patients.
